蜡蚧轮枝菌JMC-01介导的甘蓝抗桃蚜效应

摘要: 为明确蜡蚧轮枝菌JMC-01定殖甘蓝(Brassica oleracea var. capitata)后所形成共生体对桃蚜(Myzus persicae)防御作用,采用喷叶法测定JMC-01及JMC-01-甘蓝共生体室内毒力,同时观测了JMC-01对桃蚜的侵染情况,并通过转录组测序分析了JMC-01定殖后甘蓝的防御相关途径改变。结果表明:侵染5 d后,JMC-01对于成蚜室内毒力高达75%,该侵染时期内,JMC-01-甘蓝共生体对成蚜与1龄若蚜室内毒力分别为58.67%和66.67%,1龄若蚜的死亡率高于成蚜处理;使用JMC-01孢子悬浮液喷施桃蚜或桃蚜取食带菌叶片后,菌丝对虫体侵染程度不同;直接喷施孢子悬浮液,对桃蚜侵染效果和致死效果均更好;接入JMC-01后,甘蓝相关防御基因发生改变,共检测到989个差异基因;通过进一步GO富集和KEGG发现,甘蓝作用于糖基键的水解酶活性、细胞壁、光合作用-天线蛋白、芥子油苷生物合成、MAPK信号-植物通路、硫代谢等防御相关途径出现改变;JMC-01能够通过内生定殖提高植物抗性,在生物防治领域有较好应用潜力。

Abstract: Lecanicillium lecanii (JMC-01) can form symbionts after colonizing cabbage plants to defend against Myzus persicae. We tested the toxicity of JMC-01 and JMC-01-cabbage symbionts to Myzus persicae by leaf spraying method and observed the infection of JMC-01 to Myzus persicae. We further analyzed the defense-related pathway changes of cabbage plants after colonization by JMC-01 by transcriptomic sequencing. JMC-01 showed 75% virulence against adult aphids at five days post-infection, while the JMC-01-cabbage symbiont induced 58.67% and 66.67% mortality in adults and 1st-instar nymphs, respectively. Nymphal mortality was significantly higher than that of adult aphids. The infection patterns of mycelial in Myzus persicae depended on the inoculation method, either through direct spraying with JMC-01 spore suspension or via feeding on colonized leaves. Direct application of spore suspension demonstrated superior infection efficacy and mortality rates against Myzus persicae. Furthermore, cabbage plants inoculated with JMC-01 exhibited altered defense-related gene expression, with a total of 989 differentially expressed genes being identified. GO enrichment and KEGG pathway analyses revealed significant alterations in defense-related pathways, including hydrolytic enzyme activity acting on glycosyl bonds, cell wall metabolism, photosynthesis-antenna proteins, glucosinolate biosynthesis, MAPK signaling-plant pathway, and sulfur metabolism. Lecanicillium lecanii (JMC-01) can enhance plant resistance through endophytic colonization, with significant potential for application in biological control systems.
