Application of ISSR in identifying Flammulina velutipes strains.

Abstract: In this paper, ISSR molecular fingerprinting markers were employed to identify six strains of Flammulina velutipes by using 8 primers selected from 20 ISSR primers. A total of 136 DNA fragments were amplified, of which, 111 (81.6% of all bands) were polymorphic. The standard genetic identity among the test strains ranged from 0.5674 (between Nongda04-1 and Quanhe04-1) to 0.8947 (between Taihang05-1 and Hebei03-1). The UPGMA cluster analysis based on the standard genetic distances showed that cultivated F. velutipes strains could be divided into three groups. It was indicated that ISSR marker was an effective tool for the strain identification and fingerprint analysis of F. velutipes.
